Profiling YAP1 expression in breast cancer subtypes for prognostic significance

Overview

YAP1, a co-activator in gene transcription, plays an oncogenic role in various cancers, including breast cancer. Our study focuses on assessing YAP1's prognostic implications in Indian breast cancer subtypes.

Milestones
  1. Poster presentation at international conference; 42nd annual conference of the Indian Association of Cancer Research (IACR) is being held at ACTREC-TMC from the 12th to 16th of January, 2023 "Prognostic significance of YAP1 in the non-TNBC and TNBC subtype in the Indian breast cancer cohort”.

  2. Publication: Venkatasubramanian G, Kelkar DA, Mandal S, Jolly MK, Kulkarni M. Analysis of Yes-Associated Protein-1 (YAP1) Target Gene Signature to Predict Progressive Breast Cancer. Journal of Clinical Medicine. 2022; 11(7):1947. https://doi.org/10.3390/jcm11071947

Investigation of regulation of YAP driven tumorigenesis by promoter proximal pausing (PPP)

A temporary halting of RNA polymerase 2 during its elongation phase is known as promoter proximal pausing, and is found to be regulating tumorigenesis by limiting YAP1 function. We intend to investigate if PPP regulated subset of YAP targets are crucial towards neoplastic transformation in mammalian cells as well. We will be testing if patient data shows any correlation of PPP expression with YAP and patient outcomes.

Role of YAP in Growth Factor Signalling Receptor HER2 in Cancer Progression

Anti-HER2 therapies although effective, not all the patients show complete response to the therapy and such patients recur within the first five years of primary diagnosis. Our previous analysis for YAP expression across breast cancer subtypes showed a strong association with unfavourable outcomes, especially in HER2+ subtype.

Here we investigate whether YAP collaborates with HER2 in driving HER2+ breast cancer progression.
